Summary: Tributyltin(TBT) is an obesogen associated withseveralcomplications. However, fewinvestigations have evaluated TBTeffects on adult mammary glands (MG). In this investigation, we assessedwhether TBT's obesogenic effects resulted in abnormal MG fat padexpansion and other irregularities. TBT was administered to female rats,and their MG morphophysiological development was assessed. We furtherassessed the MG fat pad for PPARγ, ERα, and aromatase protein expression,as well as inflammation, oxidative stress (OS) and fibrosis. Irregular MGmorphological development such as lower TEB number, alveolar (AB), lobuleand differentiation (DF) escorewere observed in TBT rats. TBT rats hadabnormal MG fat accumulation as evidenced by increased numbers ofhypertrophic adipocytes, triglyceride (TG) levels and PPARγexpression. Astrong negative correlation between the MG obesogenic makers and TEBnumber, AB and DF escorewere observed in TBT rats. MG inflammation wasobserved in TBT rats. A positive correlation between the MG obesogenicmarkers and inflammation were observed. High ERαand aromatase expressionwere observed in MG of TBT rats. MG OS and fibrosis were present in theTBT rats. Additionally, a positive correlation between the MG obesogenicmarkers and OS were observed in TBT rats. Thus, these data suggest thatobesogenic TBT effects led to MG irregularities in the adult female rats.
