Summary: Acute kidney injury (AKI) is characterized by a rapid and potentially reversible decline in renal function; however, the current treatment of AKI is not specific and is associated with limited supportive care. Considering the need for new therapeutic approaches, we believe that the hydroalcoholic extract of leaves of Eugenia uniflora (pitanga) because of its antioxidant properties, may be beneficial for the treatment of RAI induced by renal ischemia / reperfusion (I / R).
Hydroalcoholic extract from Eugenia uniflora leaves (200mg / kg, oral) or vehicle (0.9% saline) was administered to male Wistar rats prior to the induction of bilateral renal ischemia (45 minutes). After 24 hours of reperfusion, clearances of inulin and paraminothiazurate were performed to determine the glomerular filtration rate (GFR), renal plasma flow (RPF), renal blood flow (RBF) and renal vascular resistance (RVR). The production of reactive oxygen species (ROS) and renal apoptosis were evaluated by flow cytometry. The evaluations of the expression of the antioxidant enzymes were performed by the Western blot technique. Mean ± SEM. ANOVA 2-way, Tukey post-hoc test, p <0.05.
The results showed protective effect of Eugenia uniflora extract at the dose used. Administration of the EU extract prevented I / R induced changes in GFR (SH + VH 7.5 ± 0.5 / SH + EU 6.8 ± 0.3 / I / R + VH 2.9 ± 0 , IPR + SH + VH: 16.7 ± 1.4 / SH + EU: 19.1 ± 3.3 / I / R + VH 4.6 ± 0.6 and I / R + EU 18.7 ± 2.6 mL / min / kg), FSR (SH + VH: 26.3 ± 3.0 / SH + EU: 29.3 ± 7,0 / I / R + VH: 7.6 ± 0.9 and I / R + EU: 29.1 ± 4.2 mL / min / kg) and RVR (SH + VH 4.6 ± 0, 7 / SH + EU 5.7 ± 2.0 / I / R + VH 21.8 ± 5.9 and I / R + EU 4.5 ± 1.1 au). In this study, the extract was able to prevent the increase of the production of reactive oxygen species in the animals I / R (SH + VH: 344.52 ± 23 / SH + EU: 332.2 ± 13 / I / R + VH 503.12 ± 33 and I / R + EU 370.69 ± 27 water). This prevention of the increase of ROS is related to the increase of the activity of the antioxidant enzymes SH + VH (catalase 0.710 ± 0.090 and SOD 0.27 ± 0.046); SH + EU (catalase 0.750 ± 0.110 and SOD 0.69 ± 0.020); I / R + VH (catalase 0.420 ± 0.100 and SOD 0.090 ± 0.14) and I / R + EU (catalase 0.080 ± 0.200 and SOD 0.209 ± 0.037). Moreover, the extract was able to prevent apoptosis in I / R animals (SH + VH: 4.8 ± 1.25 / SH + EU: 4.9 ± 0.19 / I / R + VH 10.20 ± 1.2 and I / R + EU 3.9 ± 0.9% , group values). These results demonstrate the protective effect of Eugenia uniflora hydroalcoholic extract on AKI.
